Officers attached to Monaro Police District have established a crime scene and the Crash Investigation Unit will also attend.
NSW Ambulance paramedics were called to the scene and treated the driver of one of the vehicles at the scene for head and spinal injuries, while the truck driver received treatment for minor injuries.
Both were taken to Canberra Hospital.
The other drivers were treated for minor injuries on scene.
The Monaro Highway is closed in both directions as of 1:30 pm on Thursday, according to Live Traffic NSW.
